Lyophilized powder of mesenchymal stem cell supernatant attenuates acute lung injury through the IL-6–p-STAT3–p63–JAG2 pathway

Abstract Background Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are syndromes of failure with extremely high mortality few effective treatments. Mesenchymal stem cells (MSCs) may reportedly contribute to tissue repair in ALI ARDS. However, applications MSCs have been restricted due safety considerations limitations terms large-scale production industrial delivery. Alternatively, the MSC secretome has considered promising for use therapeutic approaches advanced pre-clinical clinical trials. Furthermore, can be freeze-dried into a stable ready-to-use supernatant lyophilized powder (SLP) form. Currently, there no studies on role SLP ALI. Methods Intratracheal bleomycin was used induce mice, intratracheal administered as treatment. Histopathological assessment performed by hematoxylin eosin, immunohistochemistry, immunofluorescence staining. Apoptosis, inflammatory infiltration, immunological cell counts, cytokine levels, mRNA- protein-expression levels relevant targets were measured performing terminal deoxynucleotidyl transferase dUTP nick-end labeling assays, determining total protein bronchoalveolar lavage fluids, flow cytometry, multiple cytokine-detection techniques, reverse transcriptase-quantitative polymerase chain reaction western blot analysis, respectively. Results We found that considerably promoted survival, inhibited epithelial apoptosis, attenuated recruitment, reversed imbalances induced bleomycin. interleukin 6–phosphorylated signal transducer activator transcription signaling pathway activate tumor 63–jagged 2 basal cells, suppress T helper 17 differentiation, promote p63 + proliferation damage repair, attenuate responses. Conclusions ameliorated activating promoting damaged cells. The findings this study also shed insight pathogenesis imply shows considerable promise treating
